FIRST Study
Frontline Immunotherapy with Response-Guided Subsequent Treatment
This site provides supplemental materials, model-building workflows, and reproducible Bayesian analyses supporting the FIRST study, an institutional cohort study of frontline immunotherapy for high-risk cutaneous squamous cell carcinoma (CSCC).
Materials include generative modeling workflows, prior and posterior checks, convergence diagnostics, and model-specific analysis documents corresponding to the primary objective response and event-free survival analyses.
Core Workflows
Response Modeling Workflow
↗Generative Bayesian modeling for objective response, including prior specification, simulation-based validation, posterior predictive checks, and dose-response model development.
Event-Free Survival Workflow
↗Generative workflow for landmark event-free survival modeling, including prior predictive simulation, Weibull survival modeling, and sensitivity analyses.
Model-Specific Analyses
ORR Model: Linear Dose Specification
↗Full analysis for Bayesian objective response modeling under a linear dose parameterization.
ORR Model: Log-Dose Specification
↗Model-specific analysis for objective response under a log-dose formulation allowing diminishing returns.
ORR Model: Categorical Dose Specification
↗Threshold-based Bayesian objective response modeling using categorical dose indicators.
Event-Free Survival Model
↗Full model-specific analysis for the landmark Bayesian event-free survival model and associated sensitivities.
